just want to confirm for HP-UX 10.20 9000/K260 limitations, the maximum possible accessible RAM is 3.75 Gb.

Yes. The missing 0.25GB is used for device mapping. Bear in mind that you still have 1GB quadrants to deal with so your 3.75GB value is only valid with many conditions attached.

I want to confirm if I can add any RAM and CPU slots to HP10.20 9000/K260. Currently it has 3Gb and 2 CPUs.
Apart from physically checking the machine, will a shutdown for reboot then interrupt at BCH (10 seconds) to run IN ME and IN PR do the job as well? If yes, what do I need to look out for?

If you did not delete the online diagnostics, you can run this command:

echo "selclass qualifier memory;info;wait;infolog" | /usr/sbin/cstm

This will show the size of the memory modules installed. However, you may have only one memory carrier installed and I don't believe the second carrier can be identified if is installed with no RAM or simply not present.

If memory (mine, this is) serves, a K2XX could only hold 1 memory carrier. However, even in the K5xx which could hold 2 memory carrers and upto 8GB of RAM, only 4GB was usable under 10.20. In your case, with 3GB you may or may not be able to add more memory modules because not all of your modules might be 512MB. Either the online diagnostics or the firmware monitor can be used to display the memory slots and the modules in them.

my online diagnostic hasn't been updated and hence not giving the information on carrier or/and slots. Hence I understand the only way to find out is open up the machine or during reboot, type IN ME, right? In one carrier, is there 16 memory slots?

It is more accurate to say that there are 8 pairs of sockets (0a,0b .. 7a,7b) per carrier and each memory module is comprised of pair of memory boards.

You can inspect the memory carrier compartment while the machine is running. Take off the front panel cover, then remove the memory/CPU cover on the left side of the machine. You'll need Posidrive tools. NOTE: This metal cover controls ventilation so do not leave the cover off for more than few seconds. You can see the memory carrier(s) in the leftmost compartment.

As mentioned, your carrier may be full and you'll have to buy another carrier or replace a lot of the RAM modules with higher capacity modules. The K-series is very old in computer years and 10.20 has been out of support since 2002, so the expense in adding another 750 megs may not provide a meaningful improvement in performance.

The K260 only supported one memory carrier. The K460 supported two memory carriers.
