When you say Single-Mode and Multi-mode you are talking about the type of Fiber cable you are using to connect the server NIC to a switch. You also have to consider the SFP that you are using in the NIC ports. Short-Wave SFPs are normally used with Multi-mode cables for shorter distances. Long-wave SFPs are used with Single mode cables for longer distances.

So to answer your question having two different types of SFPs in the same 2-port does not make sense.
