ist it possible to setup SpamAssassin on linux and place it between firewall and mail server MS Exchange 5.5 ?
has someony any experience with SpamAssassin
in connection with MS Exchange 5.5 ?
do I need a special interface, script etc. ?

exiscan is a patch to Exim MTA for content scanning support.
you can use spam keyword in exim ACL statement "acl_smtp_mime".

I've played with things the other way, filtering mail stored pop3 mail off an exchange server.
Spam assassin does have global and individual user configuration that can reduce mail volume. Ultimately I think the best way to use it is to keep my Linux servers mailboxes from filling up.
Ultimately you have to configure the product. There are decent cookbooks on the site http://www.spamassasin.org
I'm not sure how useful the product is. It does keyword filtering which does help. email address filtering is so useless I rarely bother any more.
