Sorry, my crystal ball seems to be broken lately.
Perhaps if you told us:
* what OS
* what version of OS
* file system on boot LUN
people here just *might* have an answer.

Not only will you have to reboot, but a Google search for
windows 2003 diskpart extend system volume
it looks like you would need to:
* back up the system disk
* shut down the system
* extend the drive
* use a second system / bootable disc with diskpart (or other utility to extend the partition)
* boot the system
Looking at the links, Windows is not going to like extending the active system partition. However, if it was a data disk on *another* system, that should be a different story.
Good luck.
Don't forget the part about backing up the system disk before you start! A snapclone would be a Good Thing if you have the BC license.

Or, leave original boot LUN alone, and work on a snapclone:
1. power down the (first) host
2. take a snapclone of the boot LUN
3. allocate more space
4. Present snapclone to another (second) host
5. use diskpart to extend
6. power down second host
7. unpresent original from first host
8. present extended snapclone to first host
9. power up and check first host
If all is well, delete original boot LUN. Otherwise:
1. power down the first host
2. unpresent the extended boot LUN
3. present the original boot LUN
4. figure out what to do next....
