Can anyone enlighten me as to the differences in performance between Simplex and Duplex slots on the DL380 G4

When you duplex the drive cage, you offer the drives (depending on where you install them) the throughput of two SCSI channels vs. one SCSI channel. If you split up the arrays that you install on the server, you can gain throughput access to the disks as you can read and write to the drives over both channels instead of all 6 drives going through one SCSI channel.
