SolutionIt can be done completely on the fly. I recommend, as always, to have a backup "Just incase" though.
I recommend using the latest Array Config Utility to do this though.

I should also make note that I'm assuming you're using a Smart Array controller.

It's always good to know what exactly we are talking about - I mean a server model, controller, drives, etc.
Under normal circumstances it is not possible to create RAID 1+0 having just two drives - you need to have at least for.
However there is small catch here - Intel Matrix Storage Technology: http://www.intel.com/design/chipsets/matrixstorage_sb.htm
This allows splitting every hard drive in 2 parts & dedicating each part for different RAID level. Hence you can have 'sort of' functionality of RAID 1+0 (or rather 0+1) having only 2 drives.
But I severely doubt that adding drives on the fly is possible.
