When does the S/N of a server change?
Is it when the motherboard is replaced?

you mean SN must reinserted? I mean when you replace a SB you must enter the SN again manually, press F9 for (300/500/700 Server Series) or F10 for 100 server series
and reinsert the Serial number

only in addition : The SN of a server don't change never.
Like already explained, after Systemboard replacement it is not in the new NVRAM. It is important that you enter it, because most tools/apps read it from there...

all answers are the same as far as I see.
You have one serial number for the server - the unique HP manifacture number. It is stored in NVRAM of the server.
When you replace the systemboard you "replace"/clear the NVRAM and that is why you have to enter it again in RBSU under the serial number field.
