do you mean alignment on the client-side for a lefthand virtual disk? What client OS do you use?

Partition alignment for MBR-style partitions is 64K or a multiple.

I'm using ESX. As i red in VMFS best practise that 128k is best practise for EMC.

So 64K or 128K are both okay right?

128K is a multiple of 64K, so the answer is: Yes

but a VMFS partition is automatically aligned to 64K if it is initialized by the VI/vSphere client.

Do not forget to align the partitions within the Virtual Machines.
