Restrict traffic to single NIC

Newbie question: Customer is adding a 2nd NIC to CMS and managed servers for all monitoring traffic. How do I reconfigure the CMS and the managed systems to use ONLY this NIC/subnet ?

using private subnet (192.168.x.x), no DNS currently - will this work ?

Can this be done by editing config files, not using GUI ?
